南蛇藤提取物对人肝癌细胞增殖、迁移及血管内皮生长因子-C表达的影响Effects of Celastrus Orbiculatus Extract on the Proliferation, Migration and Vascular Endothelial Growth Factor-C Expression of Human Hepatoma Cells

摘要(Abstract):
目的探讨南蛇藤提取物抑制人肝癌转移的分子机制。方法人肝癌HCCLM6细胞生长于培养液中进行传代,取对数生长期细胞进行实验。南蛇藤提取物组共分为10、20、40、80、160mg/L5个浓度,阳性对照组给予5-氟尿嘧啶(5-Fu)注射液180mg/L,阴性对照组不做处理。分别观察南蛇藤提取物对HCCLM6细胞增殖和迁移能力的影响,并检测南蛇藤提取物对HCCLM6细胞及其培养上清中血管内皮生长因子-C(VEGF-C)蛋白表达的影响。结果南蛇藤提取物组作用24、48h时160μg/ml对细胞增殖的抑制率最高(P<0.01)。南蛇藤提取物组和阳性对照组穿膜细胞数较阴性对照组明显减少(P<0.01);南蛇藤提取物组10、20、40、80μg/ml浓度穿膜细胞数多于阳性对照组,并呈浓度依赖性(P<0.05或P<0.01)。南蛇藤提取物组各浓度和阳性对照组VEGF-C蛋白水平均低于阴性对照组(P<0.05或P<0.01)。结论南蛇藤提取物能抑制HCCLM6细胞的增殖、迁移及下调VEGF-C蛋白的表达水平,这可能是其抑制肝癌转移的部分机制。
